Genetic Analysis of Bacteriophages

NCBI Bookshelf. Modern Genetic Analysis. New York: W. Freeman; Bacteriophages, the viruses that parasitize and kill bacteria, can be used in several different types of genetic analysis. First, two distinct phage genotypes can be crossed to measure recombination and hence map the viral genome. Second, they can be used as a way of bringing bacterial genes together for linkage and other genetic studies.

Genetics of Microbes pp Cite as. The study of bacteriophages has had a crucial influence on many areas of genetics and molecular biology. Phages show great variation in structure, and examples of some of these are shown in Table 4. Unable to display preview. Download preview PDF.

Bacteria and some lower eukaryotes are unique in that they can pass DNA from one cell of the same generation to another. We refer to this as Horizontal Gene Transfer see diagram below. Conjugation is the transfer of DNA directly from one cell to another through cell-cell contact.

Metrics details. Aquaculture is the fastest growing sector of food production worldwide. However, one of the major reasons limiting its effectiveness are infectious diseases among aquatic organisms resulting in vast economic losses. Fighting such infections with chemotherapy is normally used as a rapid and effective treatment.

Horizontal Gene Transfer

Bacterial conjugation

Bacterial conjugation is the transfer of genetic material between bacterial cells by direct cell-to-cell contact or by a bridge-like connection between two cells. It is a mechanism of horizontal gene transfer as are transformation and transduction although these two other mechanisms do not involve cell-to-cell contact. Classical E. However, it is not sexual reproduction, since no exchange of gamete occurs, and indeed no generation of a new organism : instead an existing organism is transformed.

Gene mapping describes the methods used to identify the locus of a gene and the distances between genes. The essence of all genome mapping is to place a collection of molecular markers onto their respective positions on the genome. Molecular markers come in all forms. Genes can be viewed as one special type of genetic markers in the construction of genome maps, and mapped the same way as any other markers. There are two distinctive types of "Maps" used in the field of genome mapping: genetic maps and physical maps. While both maps are a collection of genetic markers and gene loci, [3] genetic maps' distances are based on the genetic linkage information, while physical maps use actual physical distances usually measured in number of base pairs.

Bacteria of the genus Pseudomonas are genetically diverse and ubiquitous in the environment. Like other bacteria, those of the genus Pseudomonas are susceptible to bacteriophages which can significantly affect their host in many ways, ranging from cell lysis to major changes in morphology and virulence. Insights into phage genomes, evolution, and functional relationships with their hosts have the potential to contribute to a broader understanding of Pseudomonas biology, and the development of novel phage therapy strategies. Here we provide a broad-based comparative and evolutionary analysis of complete Pseudomonas phage genome sequences available in online databases. Based on overall nucleotide similarity and the numbers of shared gene products, out of genome sequences were grouped into 12 different clusters; 30 were characterized as singletons, which do not have close relationships with other phage genomes.
