Print Send Add Share. Notes Abstract: Maternal nutrition can induce significant modifications in the epigenome of the fetus, altering gene expression and impacting future performance. DNA methylation, an epigenetic modification, is related to maternal nutrition through the one carbon cycle, involving aminoacid methionine. We hypothesized that methionine supplementation in beef cows during early pregnancy would impact the epigenome of the offspring, leading to changes in gene expression regulation. Muscle tissue samples were collected from the offspring at one month of age for transcriptome and methylome analysis using next-generation sequencing. A total of cytosines were found to be differentially methylated between diets, flagging differentially methylated genes involved in muscle development, calcium signaling and metabolism. Additionally, differentially methylated CpG islands were associated to genes related to development and transcriptional activity.

The extensive application of engineered nanomaterial ENM in various fields increases the possibilities of human exposure, thus imposing a huge risk of nanotoxicity. Hence, there is an urgent need for a detailed risk assessment of these ENMs in response to their toxicological profiling, predominantly in biomedical and biosensor settings. Literature relating to physicochemical characterization and toxicological behavior of ENMs were analyzed, and exposure assessment strategies were explored in order to extrapolate opportunities, challenges, and criticisms in the establishment of a baseline for the risk assessment paradigm of ENMs exposure. Various challenges, such as uncertainty in the relation of the physicochemical properties and ENM toxicity, the complexity of the dose-response relationships resulting in difficulty in its extrapolation and measurement of ENM exposure levels emerged as issues in the establishment of a traditional risk assessment. Such an appropriate risk assessment approach will provide adequate estimates of ENM exposure risks and will serve as a guideline for appropriate risk communication and management strategies aiming for the protection and the safety of humans. DNA methylation is an epigenetic modification critical to normal genome regulation and development. The vitamin folate is a key source of the one carbon group used to methylate DNA. Because normal mammalian development is dependent on DNA methylation, there is enormous interest in assessing the potential for changes in folate intake to modulate DNA methylation both as a biomarker for folate status and as a mechanistic link to developmental disorders and chronic diseases including cancer. Folate is an essential water-soluble vitamin occurring naturally in select foods as well as in the synthetic form folic acid used in supplements and in food fortification programs 1 — 3. There are many critical cellular pathways dependent on folate as a 1-carbon source including DNA, RNA, and protein methylation as well as DNA synthesis and maintenance.
